| 1. 순수분리
여러가지 미생물들이 섞여 있는 재료로부터 한 종류의 미생물을 분리하는 방법을 순수분리법이라고 한다. 이 방법에는 여러가지가 있으나 흔히 많이 사용되고 있는 방법은 한천 평판배지의 표면이나 깊숙한 내부에서 독립된 콜로니(colony)를 형성하도록 한 다음 분리하는 방법이다. 전자를 도말 평판법(streak plate method)라고 하고 후자를 주입평판법(pour plate method)라고 한다.
